I think you should - here’s why:
When promoting a server to a DC it queries DNS to find a another DC to pull a replica from. At this point, it doesn’t matter if you running dcpromo right away of if you have a backup to pre-populate AD there - it will contact another DC anyway.
When an Active Directory client starts, it tries to authenticate with the directory service in order to do further stuff like ask the DC DNS questions and download and process Group Policy. To keep traffic within reasonable borders, there’s a process built into AD that helps clients find a ‘near’ DC to query.
